The short version of deamidation fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-01-11. Anything still debated is marked as such rather than presented as settled.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.

Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
== Clinical Significance == In diseases related to the cartilage, the territorial matrix undergoes structural and material changes, leading to degradation of the joints, like in the case of osteoarthritis. Osteoarthritis occurs when the proteoglycans and collagen fibers degrade and become disorganized. Consequently, the decreased mechanical support makes chondrocytes more vulnerable to damage. Matrix-degrading enzymes including matrix metalloproteinases (MMPs) target the region which can accelerate cartilage degradation. Attempts to restore a functional territorial matrix is a particular area of interest in tissue engineering and cartilage repair, since its structure significantly contributes to optimal mechanical and biochemical behavior. Studies on biomimetic proteoglycans, which mimic the natural existing component of the ECM, and their effect on neighboring cartilage regions have shown promising results for strengthening cartilage with osteoarthritis. Additionally, biomaterials designed to mimic native composition and mechanical properties of cartilage, especially those with synthetic proteoglycans or collagen, have shown experimental promise. In combination with these structures, the durability of engineering cartilage may be enhanced by regenerating the territorial matrix and could improve integration with native tissue.
August Wilhelm joined the conservative nationalist veterans group Der Stahlhelm (English: "The Steel helmet"). In the following years he had increasing contact with the NSDAP. He joined the Nazi Party on 1 April 1930, whereupon he received the low membership number 24, for symbolic reasons. In November 1931, he was accepted into the paramilitary Sturmabteilung (SA) with the rank of Standartenführer. His involvement with the NSDAP and his adoration of Adolf Hitler made August Wilhelm often the subject of mockery by the left-wing press (who gave him the nickname Braunhemdchen Auwi, or "Auwi the Little Brown Shirt"), politicians (French Ambassador André François-Poncet called him Hanswurst "Hans the Brown Sausage") and from the National Socialists themselves (Joseph Goebbels referred to him as a "good-natured but slightly gormless boy"). As a representative of the erstwhile Hohenzollern dynasty, August Wilhelm was deliberately used by the party to gain votes in elections: he was elected as its lead candidate for the Landtag of Prussia in April 1932, and retained his seat until the Landtag's dissolution in October 1933. At the same time, he was appointed an election speaker alongside Hitler, whom he accompanied on flights across Germany. Through his appearances at the party's mass rallies, he addressed himself to sections of the population that were lukewarm towards National Socialism and convinced them "that Hitler was not a threat, but a benefactor of the German people and the German Empire".
=== EC 2.9: selenium transferases === EC 2.9 includes enzymes that transfer selenium-containing groups. This category only contains two transferases, and thus is one of the smallest categories of transferase. Selenocysteine synthase, which was first added to the classification system in 1999, converts seryl-tRNA(Sec UCA) into selenocysteyl-tRNA(Sec UCA).
==== MNPs for cosmetic and skin care ==== Skin treatment including face whitening agent and dark eye circles serum can also incorporated in MNPs. Its localized property enhance skin whitening delivery to the face area. Even a very specific spot like dark eye circles. By measuring the melanin (dark or black pigment found on the skin) index, subjects that are treated with whitening agents coated in MNPs show lower melanin index, compared to the whitening essence (topical) group. The treatment lasts for eight weeks, and the result shows MNPs might be a promising cosmetic vector because MNPs does not introduce skin irritation and can be engineered to localize or specific parts of the body.

As an AAS, ethylestrenol is an agonist of the androgen receptor (AR), similarly to androgens like testosterone and dihydrotestosterone (DHT). It has low estrogenic activity (via aromatization into ethylestradiol following transformation into norethandrolone), strong progestogenic activity, and a high ratio of anabolic to androgenic activity, similarly to other nandrolone derivatives. Like other 17α-alkylated AAS, ethylestrenol has a risk of hepatotoxicity.
Honey is a sweet and viscous substance made by several species of bees, the best-known of which are honey bees. Honey is made and stored to nourish bee colonies. Bees produce honey by gathering and then refining the sugary secretions of plants (primarily floral nectar) or the secretions of other insects, like the honeydew of aphids. This refinement takes place both within individual bees, through regurgitation and enzymatic activity, and during storage in the hive, through water evaporation that concentrates the honey's sugars until it is thick and viscous. Honey bees stockpile honey in the hive. Within the hive is a structure made from beeswax called honeycomb. The honeycomb is made up of hundreds or thousands of hexagonal cells, into which the bees regurgitate honey for storage. Other honey-producing species of bee store the substance in different structures, such as the pots made of wax and resin used by the stingless bee. Honey for human consumption is collected from wild bee colonies, or from the hives of domesticated bees. The honey produced by honey bees is the most familiar to humans, thanks to its worldwide commercial production and availability. The husbandry of bees is known as beekeeping or apiculture, with the cultivation of stingless bees usually referred to as meliponiculture. Honey is sweet because of its high concentrations of the monosaccharides fructose and glucose. It has about the same relative sweetness as sucrose (table sugar). One standard tablespoon (14 mL) of honey provides around 180 kilojoules (43 kilocalories) of food energy.
=== Research === Although bilberries have been used in traditional medicine, there are no proven health benefits or antidisease effects from consuming them. One review of low-quality clinical research concluded there was no evidence that consuming bilberries improves night vision.

== History == Originally, the name "Medicare" in the United States referred to a program providing medical care for families of people serving in the military as part of the Dependents' Medical Care Act, which was passed in 1956. President Dwight D. Eisenhower held the first White House Conference on Aging in January 1961, in which creating a health care program for social security beneficiaries was proposed. President Lyndon B. Johnson signed the Social Security Amendments on July 30, 1965, establishing both Medicare and Medicaid. Arthur E. Hess, a deputy commissioner of the Social Security Administration, was named as first director of the Bureau of Health Insurance in 1965, placing him as the first executive in charge of the Medicare program. At the time, the program provided health insurance to 19 million Americans. The Social Security Administration (SSA) became responsible for the administration of Medicare and the Social and Rehabilitation Service (SRS) became responsible for the administration of Medicaid. Both agencies were organized under what was then known as the Department of Health, Education, and Welfare (HEW), in 1965. Since then, HEW, has been reorganized as the Department of Health and Human Services (HHS) in 1980. This consequently brought Medicare and Medicaid under the jurisdiction of the HHS. In March 1977, the Health Care Financing Administration (HCFA) was established under HEW. HCFA became responsible for the coordination of Medicare and Medicaid.
==== Nomenclature ==== Papers published prior to a recent community consensus employ differing sets of nomenclature. The precursor peptide has been referred to previously as prepeptide, prepropeptide, or structural peptide. The leader peptide has been referred to as a propeptide, pro-region, or intervening region. Historical alternate terms for core peptide included propeptide, structural peptide, and toxin region (for conopeptides, specifically).
== Paregoric, U.S.P. formula == The principal active ingredient in paregoric is powdered opium. In the United States the formula for Paregoric U.S.P. is a tincture of opium 40 mL, anise oil 4 mL, benzoic acid 4 g, camphor 4 g, glycerin 40 mL, alcohol 450 mL, purified water 450 mL, diluted with alcohol to 1000 mL, and contains the equivalent of 0.4 mg/mL of anhydrous morphine; one ounce of paregoric contains 129.6 mg (2 grains) of powdered opium, or the equivalent of 13 mg of anhydrous morphine. The average adult dose is 4 mL by mouth which corresponds to 16 mg of opium, or 1.6 mg of anhydrous morphine. It is unclear when the current formula for Paregoric, U.S.P. was developed in the United States. One formula for "Camphorated Tincture of Opium (Paregoric Elixir)" attributed to the United States Pharmacoepia of 1863 is: "Macerate 1 drachm each powdered opium and benzoic acid, 1 fluid drachm of anise, 2 ounces clarified honey, and 2 scruples camphor, in 2 pints diluted alcohol for 7 days, and filter through paper." A slightly different formula is given in the 1926 pharmacoepia. The current formula for Paregoric, U.S.P. should not be assumed to be universal. For example, in the United Kingdom the formula for Paregoric, B.P. is tincture of opium 5 mL, benzoic acid 500 mg, camphor 300 mg, anise oil 0.3 mL, alcohol (60%) to 100 mL, and contains about 1/30th grain of anhydrous morphine in 60 minims, which is 25% stronger than Paregoric, U.S.P.
Metreleptin, sold under the brand name Myalept among others, is a synthetic analog of the hormone leptin used to treat various forms of dyslipidemia. It has been approved in Japan for metabolic disorders including lipodystrophy and in the United States as replacement therapy to treat the complications of leptin deficiency, in addition to diet, in patients with congenital generalized or acquired generalized lipodystrophy. The most common side effects include hypoglycemia and weight loss. It was approved for medical use in Canada in January 2024.
The antimicrobial resistance crisis also extends to the food industry, specifically with food producing animals. With an ever-increasing human population, there is constant pressure to intensify productivity in many agricultural sectors, including the production of meat as a source of protein. Antibiotics are fed to livestock to act as growth supplements, and a preventive measure to decrease the likelihood of infections. Farmers typically use antibiotics in animal feed to improve growth rates and prevent infections. However, this is illogical as antibiotics are used to treat infections and not prevent infections. 80% of antibiotic use in the U.S. is for agricultural purposes and about 70% of these are medically important. Overusing antibiotics gives the bacteria time to adapt leaving higher doses or even stronger antibiotics needed to combat the infection. Though antibiotics for growth promotion were banned throughout the EU in 2006, 40 countries still use antibiotics to promote growth. This can result in the transfer of resistant bacterial strains into the food that humans eat, causing potentially fatal transfer of disease. While the practice of using antibiotics as growth promoters does result in better yields and meat products, it is a major issue and needs to be decreased to prevent antimicrobial resistance.

Retatrutide is an investigational peptide that activates the GLP-1, GIP, and glucagon receptors at the same time. It is being studied for obesity and type 2 diabetes and has not been approved for clinical use. The internal code LY3437943 refers to the same molecule.
Most approved incretin therapies target one or two receptors, while retatrutide engages three. The addition of glucagon receptor activity is intended to raise energy expenditure. This broader mechanism is the subject of ongoing research rather than established practice.
No regulatory agency has approved retatrutide for any indication as of available public information. Development remains in later-stage trials. Availability claims found online should be treated as unverified.
It is an investigational peptide that activates three hormone receptors: GIP, GLP-1 and glucagon. It is being studied mainly for obesity and type 2 diabetes, and it is not approved for any clinical use. Published information comes from controlled trials rather than from general practice.
