Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.

Mechanistic proposals link each receptor to a different physiological role. Activation of the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors is associated with reduced appetite, slower gastric emptying, and glucose-dependent insulin release. Glucagon receptor signaling, by contrast, is associated with increased energy expenditure and altered lipid handling, though it can also raise blood glucose. The design intent is to balance these contributions so that weight reduction is enhanced without unacceptable glycemic trade-offs. How well that balance holds across individuals is not fully resolved.
Published information places retatrutide in clinical development rather than on the market as an approved therapy. Early-stage and mid-stage trials have examined tolerability and changes in body weight, and larger studies continue to report results over time. Open questions include the durability of effects after treatment stops, the composition of weight lost, and cardiovascular outcomes over long periods. Statements about definitive benefit should therefore be treated as provisional. Regulatory status varies by jurisdiction and changes as applications are reviewed.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Organ transplant recipients receive immunosuppressive therapy that is associated with an increased risk to develop skin cancer, so they are advised to avoid sunlight exposure, and to take a vitamin D supplement.
California, Illinois, and New York passed the first mandatory minimum sentences for drug offenses; Congress followed with the Boggs Act of 1951, creating the first federal mandatory minimums for drugs. The act unified penalties for the Narcotic Drugs Import and Export Act and the Marihuana Tax Act, effectively criminalizing cannabis. Anslinger testified in favor of the inclusion of cannabis, describing a "stepping-stone" path leading from cannabis to harder drugs and crime. First-offense possession of cannabis carried a 2–10-year minimum and a fine of up to $20,000. This marked a change in Congress's approach to mandatory minimums, increasing their number, severity, and the crimes they covered. According to the United States Sentencing Commission, reporting in 2012: "Before 1951, mandatory minimum penalties typically punished offenses concerning treason, murder, piracy, rape, slave trafficking, internal revenue collection, and counterfeiting. Today, the majority of convictions under statutes carrying mandatory minimum penalties relate to controlled substances, firearms, identity theft, and child sex offenses.". In 1961, the Single Convention on Narcotic Drugs became the first of three UN treaties that together form the legal framework for international drug control, and require that domestic drug laws in member countries comply with the conventions. The Single Convention unified existing international drug agreements, and limited possession and use of opiates, cannabis and cocaine to "medicinal and scientific purposes", prohibiting recreational use.

== Types of bar == The majority of food bars are sweet, rather than savory. Different types of bar may be marketed for specific purposes, like replacing breakfast or providing protein, or may be marketed to specific demographics like women. Some bars are made to adhere to dietary restrictions like the paleolithic diet or the gluten-free diet.
==== Mount Koya ==== It was developed by Mokujiki Shonin, a Shingon priest in the early 1600s during the Edo Period. He encouraged all the mountain temples to make their own tofu after he obtained large amounts of soybeans from the head Shingon temple. The purpose of this was to preserve some of the frozen tofu until the Spring equinox. Firm tofu was left outdoors in the cold windy night to freeze. After it was frozen, it was allowed to stand on shelves in a shed for fifteen days at temperatures below freezing, thawed in warm water and pressed lightly to expel the melted ice, then dried in the shed using heat from charcoal braziers.

== Applications == Because of its ability to cause chemical reactions and excite fluorescence in materials, ultraviolet radiation has a number of applications. The following table gives some uses of specific wavelength bands in the UV spectrum.
Badly decomposing bodies, trauma cases, frozen, or drowned bodies, and those to be transported over long distances also require special treatment beyond that for the "normal" case. The restoration of bodies and features damaged by accident or disease is commonly called restorative art or demisurgery, and all qualified embalmers have some degree of training and practice in it. For such cases, the benefit of embalming is startlingly apparent. Normally, a better result can be achieved when a photograph and the decedent's regular make-up (if worn) are available to help make the deceased appear more as they did when alive. Embalming autopsy cases differs from standard embalming because the nature of the post-mortem examination irrevocably disrupts the circulatory system, due to the removal of the organs and viscera. In these cases, a six-point injection is made through the two iliac or femoral arteries, subclavian or axillary vessels, and common carotids, with the viscera treated separately with cavity fluid or a special embalming powder in a viscera bag. Long-term preservation requires different techniques, such as using stronger preservatives and multiple injection sites to ensure thorough saturation of body tissues.
During the Islamic Golden Age under the Abbasid Caliphate, prominent Muslim physicians preserved, systematized and developed ancient medical knowledge from across the Eurasian continent. They synthesized concepts from classical antiquity (see: Ancient Greece, Ancient Rome), Persia, Ayurveda, and China. This work laid the foundations for later advances in medieval European medicine as European physicians came into contact with Islamic authors through the Renaissance of the 12th century. Rhazes (c. 865–925), or Muhammad ibn Zakariya al-Razi, included writings about diabetes in the more than 230 books he produced in his lifetime. Avicenna (980–1037), or Ibn Sina, was a court physician to the caliphs of Baghdad and a key figure in medicine who compiled an exhaustive medical encyclopedia titled The Canon of Medicine. His account detailed the clinical features of diabetes, and termed the disease albulab ("water wheel") and zalkh el kuliah ("diarrhea of the kidneys"). He documented "the abnormal appetite and the collapse of sexual functions" and the sweet taste of diabetic urine, and further differentiated diabetes associated with emaciation from other causes of polyuria. He also elaborated on diabetic gangrene and treated diabetes using a mixture of lupine, trigonella (fenugreek), and zedoary seed. It has been noted that references to diabetes expanded in the medical texts of this period. Eknoyan and Nagy (2005) speculate that this indicates the increasing prevalence of the disease.
Hyperthyroidism (an example is Graves' disease) is the clinical syndrome caused by an excess of circulating free thyroxine, free triiodothyronine, or both. It is a common disorder that affects approximately 2% of women and 0.2% of men. Thyrotoxicosis is often used interchangeably with hyperthyroidism, but there are subtle differences. Although thyrotoxicosis also refers to an increase in circulating thyroid hormones, it can be caused by the intake of thyroxine tablets or by an over-active thyroid, whereas hyperthyroidism refers solely to an over-active thyroid. Hypothyroidism (an example is Hashimoto's thyroiditis) is the case where there is a deficiency of thyroxine, triiodothyronine, or both. Clinical depression can sometimes be caused by hypothyroidism, whereas bipolar disorder can sometimes be caused by hyperthyroidism. Some research has shown that T3 is found in the junctions of synapses, and regulates the amounts and activity of serotonin, norepinephrine, and γ-aminobutyric acid (GABA) in the brain. Hair loss can sometimes be attributed to a malfunction of T3 and T4. Normal hair growth cycle may be affected disrupting the hair growth. Both thyroid excess and deficiency can cause cardiovascular disorders or make preexisting conditions worse. The link between excess and deficiency of thyroid hormone on conditions like arrhythmias, heart failure, and atherosclerotic vascular diseases, have been established for nearly 200 years.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
